Background
The fusion gene, formed by chromosomal translocation and rearrangement, is the result of both genetic and environmental factors. Fusion genes occur at different frequencies in different ethnic groups, so fusion gene detection is one of the main bases for the diagnosis typing of some leukemias. Moreover, fusion genes are also major molecular markers for prognostic stratification, minimal residual disease (MRD) monitoring, and targeted therapy in leukemia patients.
RT-PCR is able to detect fusion genes resulting from structural chromosomal variations. By optimizing specific primers, 16 parallel multiplex PCR reactions are set up to simultaneously detect 177 mRNA fusion gene subtypes, greatly reducing the operating process.

Detection Process
In this method, RNA is isolated and purified from leukocytes in peripheral blood or bone marrow samples, and cDNA is obtained by reverse transcription to detect 177 mRNA fusion gene subtypes in 58 fusion genes that may occur in ALL, AML, MDS, MPN and other blood diseases by multiplex reverse transcription-polymerase chain reaction (RT-PCR).
