Background
The human leukocyte antigen (HLA) system (the major histocompatibility complexMajor Histocompatibility Complex [MHC] in humans), is an important part of the immune system. The cell surface antigen encoded by HLA has become an indistinguishable "feature" of everyone's cells and is the basis for the immune system to distinguish itself from foreign substances.
HLA is the most complex genetic polymorphism known to humans and can be classified as HLA-I, HLA-II, and HLA-III molecules. For HLA genotyping, it is mainly aimed at HLA-I and HLA-II. In bone marrow and organ transplantation, the severity of graft rejection is largely influenced by the level of HLA compatibility between donors and recipients. Therefore, a donor with highly identical HLA is the key to successful bone marrow and organ transplantation. Moreover, specific HLA genotypes have been shown to have a strong association with particular diseases or to trigger certain diseases. Therefore, HLA genotyping can be effectively utilized for differential diagnosis of such diseases.
Clinical Significance
High resolution genotyping can improve the matching accuracy, ensure the success rate of transplantation, and reduce the risk of transplant rejection. This can lead to a faster recovery and a longer survival period for patients post-operation.
Target Population
First-time patients with acute leukemia, chronic myelocytic leukemia, malignant lymphoma, multiple myeloma, myelodysplastic syndrome, and severe aplastic anemia; patients/donors requiring HSCT or organ transplant.

Specimen Requirements
Peripheral blood/bone marrow, 3–5 mL EDTA anticoagulation, 4 °C, within 48 h for testing.
